Title: The use of chemometric methods to investigate interactions between selected groups of drugs and food + 2 Annexes

Abstract:

Food-drug interactions (FDIs) are underexplored compared to drug-drug interactions, despite their potential impact on pharmacotherapy safety and efficacy. The dissertation investigated FDIs for selected drug groups using a two-step approach. In the first step, systematic reviews were performed to evaluate the effect of dietary interventions on the pharmacokinetics of levothyroxine, proton pump inhibitors, medications for osteoporosis, Parkinson's disease, and anti-infective agents; and evidence-based recommendations were developed for their use with food, beverages, antacids, and mineral supplements. For anti- infective agents (antiretrovirals, antibacterials, and antifungals), the efficient data was collected for a second step - multivariate analyses using chemometric techniques (e.g., principal component analysis, partial least squares modeling, and cluster analysis). Significant correlations were revealed between drug structure (expressed by molecular descriptors) and changes in key pharmacokinetic parameters (AUC, Cmax, Tmax) following dietary interventions. The research underscores the clinical impact of FDIs and demonstrates the innovative potential of chemometric methods to optimize anti-infective therapy, suggesting their possible utility in drug development to predict and mitigate FDIs.
